From what I remember of the Man-Thing origin he was working on the super serum that made Captain America, he wife betrayed him and I think the Mafia, came in and either gun him down and set fire to the place or just blew the place up, and the serum mixed with the swamp, and changed Ted Sallis.

EDIT: To keep AIM from getting its hands on the serum he devised to allow people to survive in polluted environimants, chemist Ted Sallis injects himself with it, and its properties combine with the mystic powers of the swamp to turn him into a near-mindless Man-Thing.

I was way off.
