I'm 6 yrs older than my brother. We got along really well. He would get protective of me at times but mostly his guy friends who liked me would check it with him first. Half the time he'd be the one to ask me if I'd go out with them - being the matchmaker sortof. If he didn't like them though they never asked me even though he'd tell me that they liked me. He didn't mind his friends flirting with me but he knew that if he didn't think the guy would be good for me then I probably wouldn't do anything more than flirt. He was a pretty good judge of whether I'd end up really liking the guy or not.
I'd suggest not pursuing anything with her if you value your friendship. If he didn't mind and you made sure he knew that you wouldn't go any further you could flirt with her but I would make sure he knew that was all it was.
Little brother's or big are so often protective of their sisters.
