The test is to check your iron levels to make sure you have enough that it won't endanger you to give blood.
The iodine they use (if you're not allergic) is to cleanse the site where they'll take the blood.
I used to donate regularly but my hemoglobin (iron) levels haven't been high enough for the last year or so.
