Assuming you want to go stealth:
The voice is by far the single most difficult part of passing, and it can't be mastered in a day. Mannerisms and walk are second most difficult, and they, likewise can't be mastered in a day.
Appearance is easier. Go subtle on the makeup; use just enough to make yourself look feminine without going overboard. A good even foundation is most important, with a little subtle lip color. You're less likely to be read if you tuch well and wear a nice pair of women's slacks and a blouse than if you wear a dress or skirt. I know that sounds counter-intiuitive, but it is nonetheless true. Get some shoes with a bit of a heel, and practice walking in them, both to get used to them and to break them in.
If you're just interested in glamming it up then forget the above and just go overboard with everything. Depends entirely upon which route you're taking.
