I liked this movie very much, and am happy to see the roles Bill Murray has been taking on these past few years. He's showing that comedy can be real, not contrived.
I also agree with willravel on Defoe's performance. Just his appearance onscreen in this movie was hilarious. He's good at comedy and I'd like to see him do more.
On a side note, when my wife and I were leaving the theater, we were walking behind 2 guys who had just seen it also. Their conversation went a bit like this:
Spoiler: guy1: That movie sucked.
guy2: Yeah it did. The whole movie was about getting that shark, and he didn't even kill it.
guy1: Yeah, that was stupid. Bill Murray used to be funny. Now he's making suck-ass movies.
guy2: Yeah, why can't he go back to making good movies like "Stripes" or something like that?
My wife and I just looked at each other. They just didn't get it.
